鋅對生長(cháng)期大鼠海馬Uch2L1 表達的調控作用

摘要:目的 探討鋅對生長(cháng)期大鼠海馬Uch2L1 表達水平的調控作用。方法 雄性斷乳Wistar 大鼠36只,按體重隨機分為對照組(ZA) 、缺鋅組(ZD) 和缺鋅對喂組(PF) ,ZD 組喂飼含鋅量115mgPkg 的缺鋅飼料,PF組和ZA 組喂飼含鋅量30mgPkg 的足鋅飼料,PF 組攝食量與ZD 組一致。喂飼4 周后,測定血漿鋅含量和堿性磷酸酶活性以評價(jià)機體鋅狀況,用避暗試驗測定各組大鼠的學(xué)習記憶能力,采用Western blot 和RT2PCR 方法檢測各組大鼠海馬Uch2L1 蛋白及其mRNA 的表達水平。結果 ZD 組血漿鋅含量及堿性磷酸酶活性均顯著(zhù)低于PF 組和ZA 組;ZD 組大鼠在避暗試驗中的進(jìn)洞潛伏期顯著(zhù)低于PF 組和ZA 組;與PF 組和ZA 組相比,ZD組大鼠海馬Uch2L1 及其mRNA 表達水平明顯降低。結論 缺鋅可下調大鼠海馬Uch2L1 表達水平。

Study of zinc on the modulation of Uch2L1 expression in hippocampus of growing rats

Abstract : Objective  To investigate the effect of zinc on Uch2L1 expression in hippocampus of growing rats.Methods  Thirty2six male weanling Wistar rats were randomly assigned to zinc2adequate (ZA) , zinc2deficient (ZD) and pair2fed (PF) groups. Rats of ZD group were given free access to a basal diet (Zn 115mgPkg) . Rats of other two groups were given the basal diet supplemented with zinc sulfate ( Zn 30mgPkg) . PF rats consumed the same intakes as the corresponding ZD partner on the previous day. After 42wk treatment , plasma zinc concentration and alkaline phosphatase activity were tested. Passive avoidance performance was used to examine learning and memory function. Western blot and RT2PCR were used to assess the expressions of Uch2L1 protein and mRNA in rat hippocampus , respectively. Results  Zinc deprivation dramatically decreased plasma zinc concentration and alkaline phosphatase activity. Significantly shorter latency
period in passive avoidance performance in ZD group were observed. Uch2L1 protein and mRNA expression levels in hippocampus significantly reduced in comparison with ZA and PF groups. Conclusion  Zinc deficiency could induce
downregulation of Uch2L1 expression.
